I am curious about Infect(poison counters) and phases

When is the amount of poison counters a player has checked? I have a deck that uses a t2 infect combo, but I would like to know in which phase the amount of poison counters is checked.

It's a state-based action check. Basically, it's checked as soon as they get a poison a counter and before anyone gets priority. Similar to their life being 0 after damage.

104.3d If a player has ten or more poison counters, he or she loses the game the next time a player would receive priority. (This is a state-based effect. See rule 704.)

704.1. State-based actions are game actions that happen automatically whenever certain conditions (listed below) are met. State-based actions dont use the stack.

More precisely, the game checks for state based actions whenever a player would get priority. This will almost always be right after the distribution of poison counters, but not necessarily. Adding poison counters does not trigger the state-based action.

Okay so my combo goes

T1 Glistener Elf or Inkmoth Nexus

T2: Blazing Shoal discarding Progenitus or Reaper King onto one of the creatures. If I am attacking with Glistener Elf I also play Berserk before I deal combat damage.

Right when I deal CB Damage the game is over?
