Can an indestructible creature die from a combination of damage and toughness decreasing enchantments?

If I cast Ensoul Artifact on my Darksteel Citadel and attack, and it is blocked by my opponent's Felhide Minotaur after which my opponent casts Ulcerate on the Citadel, does the citadel die?

If yes, would it still die if he had cast Ulcerate first?

No. Indestructible objects cannot be destroyed by lethal damage or by destroy effects. That's literally what "indestructible" means.

Darksteel Citadel will be a 5/5 with 2 damage marked on it after combat damage is dealt. It will be a 2/2 with 2 damage marked on it after Ulcerate resolves. It will still be indestructible, so the 2 damage can't destroy it.

Remember also that damage does not reduce toughness.
