What is the interaction between Trinisphere and Lodestone Golem?

Just as the question asks. If I have a Trinisphere and a Lodestone Golem on the field and my opponent wants to cast, say, a Brainstorm how much would the brainstorm cost?

Would it cost three or four mana?

Three. Figuring out the cost for a spell goes in a specific order.

(Cost OR Alternate cost) + (Additional Costs) - (Cost Reductions), then cost-setting effects like Trinisphere .

So, a Brainstorm would cost (U) + (1) - (0) = 1U --> bring to 3 mana --> 2U.

And, for what it's worth, from Gatherer:

Trinisphere's ability affects the total cost of the spell. It is applied after any other cost increasers or cost reducers are applied: First apply any cost increases. Next apply any cost reducers. Finally look at the amount of mana you have to pay. If it's less than three mana, you'll pay three mana.
