How helpful is Hexproof exactly?

Let's say I have a really nice creature on the field, say Griselbrand , and my opponent starts to find him to be a nuisance, so he decides to Go for the Throat and be done with him. But what if I cast Sheltering Word on Griselbrand and give him Hexproof. Would Go for the Throat resolve, and I lose my precious demon, or does he live to kill another day?

When a permanent has hexproof to start with, it prevents your opponent from targeting it at all but allows you to still attach auras or use pump spells like Giant Growth. Giving it hexproof at instant speed basically "deflects" an enemy's spell that targets that creature. Even though they targeted it at one point, the spell cant resolve with the desired effect if it no longer has a legal target. So as Epochalyptik said, the Go for the Throat will resolve, but will not kill Griselbrand because it was no longer a legal target.
"608.2b If the spell or ability specifies targets, it checks whether the targets are still legal. A target that's no longer in the zone it was in when it was targeted is illegal. Other changes to the game state may cause a target to no longer be legal; for example, its characteristics may have changed (in this case you gave it hexproof) or an effect may have changed the text of the spell. "

The end result is a countered Go for the Throat and a surviving Griselbrand .
If you give Griselbrand hexproof while Go for the Throat is still on the stack, Griselbrand becomes an illegal target for Go for the Throat and Go for the Throat will then fizzle.
