Does it effect indestructible creatures and can effectively make them die anyway?

Indestructible stops something from being "destroyed", and only two things in the game "destroy":
- Effects that specifically say "destroy" (Murderous Cut, End Hostilities, etc.)
- Lethal damage marked on a creature - either damage greater than or equal to its current toughness or any damage from a source with deathtouch
Having 0 or less toughness will cause a creature to die, but it isn't a "destroy" effect. Death Wind and other similar effects can be used to kill an indestructible creature.

Yes. Any time a creature's toughness is reduced to 0 it goes to the graveyard as a state-based action. Indestructibility does not prevent this.
