How trample works is that you must assign lethal damage to all blockers, and any damage left over "tramples over" to the defending player. If Prophetic Flamespeaker is hit by Giant Growth to make it a 4/6 and is blocked by Llanowar Elves the first strike damage will deal 4 damage, assigning 1 to Llanowar Elves which is lethal and the remaining 3 damage tramples over. State based actions send Llanowar Elves to the graveyard, and while Prophetic Flamespeaker is still "blocked" there are no blockers, so it deals 4 more damage to the defending player when non-first strike damage occurs.
