Say I have Sarkhan, the Dragonspeaker on the field, and use his +1.

Can an opponent respond to the activation of this ability and cast Hero's Downfall before Sarkhan's ability resolves and he becomes indestructible?

Yes. Loyalty abilities are activated abilities. They have to use the stack. Therefore, your opponent can respond to them. What your opponent cannot do is respond at a time after you've announce the ability and before the number of loyalty counters on the planeswalker changes.

Hero's Downfall doesn't have this problem because it outright destroys the planeswalker, but a burn spell might not be able to remove a planeswalker for that reason.
