priest of penance-pestilence

So this is a casual deck I just made: esper penance but I have one question. Lets say I only have one creature on the battlefield, High Priest of Penance enchanted for example with Hero's Resolve. currently my High Priest of Penance is 2-6. My opponent has 10 creatures on the battlefield. Normally 6 activations of Pestilence would deal lethal damage to my High Priest of Penance but I would destroy 6 nonland permanents of my opponent right?Now my question is can I trigger more times the ability of Pestilence in response to the sixth activation to destroy more creatures of my opponent?

please help me out here+tell me your opinion about the deck taking into consideration its a budget, casual deck

Each activation of Pestilence is separate. Once you you reach your High Priest of Penance's total toughness it dies as a state based action and can not longer be the target of Pestilence.

Once the High Priest of Penance in your example has 6 damage marked on him, he'll be destroyed the next time State-Based Actions (SBAs) are checked, which is right before the next time a player would get priority. You won't get an opportunity to hit him for more damage.

Why can't I activate in response to another activation? I thought I was wrong but I dont see the reason. I pay the cost(swamp) and the ability goes on stack, in response to that I activate it again, no?

Yeah, if there are more Pestilence triggers waiting on the stack at the time Priest is actually dealt the 6th point of damage, it will be dead before the next one of them resolves.
