Melira, Sylvok Outcast and Persist

If you have a Melira, Sylvok Outcast, does a card like Glen Elendra Archmage become basically immortal?

You would be correct. Persist only cares if there's a -1/-1 counter on the creature when it dies. If there's not, the creature comes back with a -1/-1 counter. Melira prevents the counter from being placed, so it enters instead with no counters on it.
