So my question if a creature with persit comes back from the graveyard with a minus one counter on it if i put a plus one counter on it do the counters cancel each other out and get removed from the creature allowing me to do it again Ok so my deck. Is called quillpod and the cards that have the interaction are Safehold Elite and Ivy Lane Denizen

Yes. If a permanent has N +1/+1 counters and N -1/-1 counters on it, those counters are removed as a state-based action. This allows you to reuse persist/undying effects if you can properly nullify the counters.

Right from the rules book:
121.3. If a permanent has both a +1/+1 counter and a -1/-1 counter on it, N +1/+1 and N -1/-1 counters are removed from it as a state-based action, where N is the smaller of the number of +1/+1 and -1/-1 counters on it. See rule 704.

Yes, +1/+1 counters and -1/-1 counters cancel each other out.
