If both Vito, Thorn of the Dusk Rose and Conclave Mentor die at the same time, will the life gain effect from Conclave Mentor still cause Vito, Thorn of the Dusk Rose effect to trigger and make my opponent lose life?

No, Vito has to be on the field when the trigger from Mentor resolves.
