if my oponent cast a Twinflame on one of his creatures and i Doom Blade it in response, does he get a copy until end of turn or not?

No, when Twinflame resolves there is no legal target

More specifically: If, as a spell starts trying to resolve, all of its targets are no longer legal, the spell is countered by the game rules.
