Do the +1/+1 counters nullify -1/-1 counters?

If i have a creature with persist and a -1/-1 counter on it andi put a +1/+1 counter can i trigger the persist one more time?

Yes. A +1/+1 counter and a -1/-1 counter on the same object will remove each other as a state-based-action.

In the case of persist and undying, it's worth noting that if a creature dies as a result of -1/-1 counters being placed onto it, the counters will be canceled out at the same time the creature dies, so the creature's last known information (the state in which it last existed on the battlefield) will show that it had both kinds of counters, and neither ability will trigger for it.
