How many times per turn can a creature regenerate?

For a example a creature with regenerate blocks a creature with power equal to its toughness. Then I pay the mana cost to regenerate, but then they do Lightning Strike on the same defending creature but I have enough mana to pay the regenerate cost, can I regenerate again?

Note that regeneration shields may be added ad nauseam, but they are consumed one at a time. They must exist before the permanent would be destroyed.
Also note that a creature is removed from combat when it is regenerated (that is, when the regeneration shield is consumed, not when it is created). Assuming Lightning Strike would kill your blocker on its own, if your opponent casts Lightning Strike at the end of the declare blockers step, your creature will be removed from combat before it can either deal or receive damage. The original activation of its regeneration ability will still resolve and create a regeneration shield, but that shield won't be necessary because your creature is out of combat and won't be hit by the attacking creature.

Sorry for all the questions, I have alot, can a creature with exploit sacrifice a creature with regenerate, then you pay the regenerate cost, but still get the benefit of the exploit effect.

No. Regeneration only replaces destruction events, which are caused by lethal damage to creatures or by effects that specifically say "destroy." Sacrifice is neither of those, so regeneration does not apply to it.

Thanks for all your answers, my brother says to regenerate means to tap the card and remove it from combat, if I block (combat phase) with a creature that has regenerate then pay the cost, then after its been tapped I can't regenerate again if Lightning Strike (an any time instant) was cast during my second main phase against that creature because it can't be tapped (it already is tapped) and removed from combat.

That is incorrect. Being tapped has nothing to do with whether an ability can be activated (the obvious exceptions being abilities that require tapping as part of their costs). Further, being tapped has nothing to do with whether regeneration shields can be created or consumed, and regeneration functions at all times (not just in combat).

Regenerating is usually an activated ability (written as "Cost : Effect"), and there's no limit to how many times you can activate an ability as long as you're able to pay for it. In your example it's fine to regenerate the creature again.
