All the werewolves on their human side check during each upkeep, including your any opponents upkeeps, for how many spells were cast LAST turn. If it was none they will transform. All werewolves on the wolf side are going to tramsform when the second spell by the same player in a turn is put on the stack.

The trigger on the opponents turn after you cast him looks at your last turn and sees at least one spell you cast so no flippies. If you do cast two instants on opponents turn and he is already wolfish he transforms back right then.

Werewolves do not transform as soon as a player casts his or her second spell of the turn. Both transform abilities trigger at the beginning of each upkeep if their condition is met.
