Grim affliction

If I play Virulent Wound and then Grim Affliction , does my opponent get an infect counter if the proliferate kills it?

Yes. It doesn't matter when the creature goes into the graveyard during that turn because Virulent Wound 's "if" ability happens when the creature hits the grave, it doesn't check to see if the creature hit the grave before a certain phase, it triggers exactly when the creature hits the grave during that turn. Grim Affliction would resolve and send the creature to the grave, when that happens Virulent Wound 's "if" ability would trigger, giving your opponent a poison counter.

on top of the above answer if your opponent doesn't already have a poison counter you will not be able to prolif the poison counter from virulent wound.

Ok, so does my opponent get 1 or 2 poison counters. This is the part that confuses me. Sorry for not adding this in the question.

Short Answer:

The only way you could give your opponent 2 poison counters with this combo is if your opponent already had a poison counter.

Reasoning:

Your opponents 2 or 3 toughness creature will die due to having 0 or less toughness as a state-based effect. State-based effects are checked right before a player would receive priority, and no player receives priority during the resolution of a spell. This means that your opponent will not get a poison counter until after the proliferate resolves. If there is no other poison counter to proliferate, then you cannot choose the poison counter that your will get after you've proliferated.
