Can you still cast an Instant after declaring blockers?

example: after my opponent declared blockers, can I cast Lightning Axe and target a designated blocker? if that will kill the targeted creature, will it deal direct combat damage?

In order to move to the next step or phase, players need to pass priority.

When the declare blockers step begins, the defending player declares blockers as a turn-based action. This process doesn't use the stack. Players then get priority to cast spells or activate abilities before the game moves to the combat damage step. So yes, you do have an opportunity to cast Lightning Axe.

However, once an attacking creature is blocked, it remains blocked for the rest of combat. Even if you remove the blocker, it won't deal damage to the defending player or planeswalker unless it has trample.

Yes, you can cast instants after blockers are declared, but before combat damage is assigned. Blockers are declared as a turn based action during the Declare Blockers step. After all blocks have been declared and ordered, there is a round of priority starting with the active player (whose turn it is). If you wish to cast a spell before damage, this is the last opportunity. If a creature is removed from combat (due to lethal damage from Lightning Axe or some other effect), it does not deal any combat damage.
