Blocking a trampler

If I block a large trampling creature with Favored Hoplite and trigger his heroic with an instant! can my opponent still deal trample damage over the top?

Yes.

When a creature (including one with trample) damage prevention effects, including protection, as well as damage increasing effects (such as Dictate of the Twin Gods ) are not taken into account. The trampler will only need to assign the Favored Hoplite lethal damage before assigning the rest to the player or planeswalker being attacked (assuming there are no other blockers); "lethal damage" is considered to be (and I'm very much paraphrasing here) "damage which, when added to the damage already marked on a creature, would cause the damage marked on that creature to equal or exceed its toughness."

If the Hoplite is a 2/3 with the damage prevention up and it blocks a 5/5 trampler, its controller could still be assigned 2 damage.
