Butcher's Glee clarity

So if I attack with a 3/3 and they declare a 1/4 Blocker and I cast Butcher's Glee on my 3/3 it will still kill the blocking creature because the regenerate would never kick in as it is a "shield" effect only triggered IF death was imminent correct?

Correct, although regeneration is not triggered.

Regeneration is a replacement effect that is applied instead of the next destruction event that would happen to the regenerated permanent. Regenerating something places a "regeneration shield" on it, and the shield is only consumed the next time that permanent would be destroyed in the current turn. The act of creating the shield does not cause it to be immediately applied.

Now, what this all means is that the regenerated creature won't be removed from combat here because it wouldn't be destroyed. It will deal 6 lifelink damage to the blocker, and the blocker will deal 1 damage back.
