Can Ajani, Mentor of Heroes use his first ability to put a +1/+1 counter on Soldier of the Pantheon ?

No. Soldier of the Pantheon has protection from multicolored. Part of protection is that you cannot target the protected object with spells or abilities of the specified quality. Ajani, Mentor of Heroes is multicolored, so its abilities can't target Soldier of the Pantheon .

Nope. Remember the acronym, DEBT. Damaged, Enchanted, Blocked, Targeted. In this instance the ability originates from a multicolored source, Ajani, Mentor of Heroes and cannot choose Soldier of the Pantheon as a legal target.
