Does Tuskguard Captain work if you have a creature with multiple +1/+1 ?

Say I have a creature with 4 +1/+1 counters would Tuskguard Captain's ability still work on it?

The ability just checks whether any +1/+1 counters are on your creatures, not the exact number of counters on each creature that has counters on it.
"Each creature you control with a +1/+1 counter on it" means "Each creature you control with at least one +1/+1 counter on it."

Yes, only creatures with 0 +1/+1 counters will miss out on the trample Tuskguard is handing out.
