Azor's Elocutors' first ability triggers at upkeep to put a counter on it, then checks if there are 5 or more counters, and if there are, wins the game. This is a single ability that happens on your upkeep and no player gets priority between the two events.
It only checks for a win condition, of 5 or more filibuster counters, during the resolution if the upkeep trigger. It doesn't check if it gets counters at any other times.
