When multiple creatures block a creature with first strike, does the first strike apply when damaging all of them?

Yes.

What happens when a creature with first strike or double strike is involved in combat is that two damage steps are used instead of one. Combat looks like this:

  • Beginning of Combat step

  • Declare Attackers step

  • Declare Blockers step

  • First Combat Damage step: during this damage step, only creatures with first strike or double strike will deal damage, no matter how many creatures are blocking or being blocked.

  • Second Combat Damage step: during this damage step, only creatures without first strike deal damage (double strike or neither ability), if they're still alive.

  • End of Combat step.

So if you attack with a 3/3 first strike and your opponent blocks with three 1/1s, then your 3/3 will deal damage to each of them during the first damage step, but they won't deal damage back because they don't have first strike or double strike. Your creature will not deal damage during the second damage step because it doesn't have double strike.

If you attack with a 3/3 first strike and your opponent blocks with two 2/2s, then you choose what order your 3/3 deals damage to them during the first damage step. One will die, but the other will survive, and deal 2 damage back to your creature during the second damage step (your creature will not deal damage due to having first strike, not double strike).

And so on.
