If I cast Asphyxiate on my opponent's untapped Wirewood Elf , can he tap for mana to save the creature?

Yes. Your opponent can tap Wirewood Elf in response to Asphyxiate to cause it to fizzle. When Asphyxiate goes to resolve it checks to see it it has a legal target, including whether the creature is untapped, and if it is a valid target the spell will resolve.
