Outland Colossus’s Second Ability

Outland Colossuss second Ability prevents him from being blocked by more than one creature. Now, what if one were to enchant him with the opposite, say an effect like the one found on Madcap Skills? They arent entirely conflicting so, do you play by the most recent effect to enter the field like usual, or does he become unblockable?

It effectively becomes unblockable. There are two restrictions in your example on how the defending player is allowed to block the Colossus: "Can't be blocked my more than one creature" and "Can't be blocked except by two or more creature". The only legal choice under those circumstances is "no blocks".
