Hexproof in Response

Can I use Blossoming Defense in response to a Doom Blade to save my creature? Or does hexproof only work before they targeted my creature since it already became the target of a spell?

Most definitely. The Blossoming Defense would trigger first, then the Doom Blade would have an illegal target and fall to the wayside. Magic follows the LIFO rule (Last In, First Out) for the Stack.
