Does Palisade Giant prevent all combat damage?

does Palisade Giant take all damage even if it goes over his toughness? For example if my opponent attackā€™s with 100 1/1s does she take all the damage or do I take 100-toughness?

Yes.

Combat Damage is dealt simultaneously and you can apply the replacement effect of Palisade Giant to that single event of damage. After the damage, state-based actions will check to see if any creatures have damage marked on them more than their toughness and find the giant with 100 damage marked on it, and propmtly the giant will die.

All damage dealt at one time goes to the giant. If some of the creatures deal damage prior to the others due to first strike or double strike, then the giant must survive that damage in order for it to take the regular combat step damage.
