Can you give a creature trample, if it already has it?

For example: Soulbright Flamekin using its ability twice on a creature.

Sure can. Having trample multiple times is redundant (it doesn't stack for additional effect), but you can certainly target the same creature multiple times with the Flamekine in order to get the eight mana boost.

No, it will only add the mana on the 3rd activation, not on any subsequent activations. You check out the Gatherer rulings here, which is generally the easiest place to get info like this.

No, because after the third time the ability resolves, you won't be able to add any additional mana because it will be after "the third time". (You can still, however, give as many creatures trample as you wish.)
