Can I respond to a spell targeting my creature with my Vines of Vastwood?

Yes. The effect targeting your creature would go on the stack, so you can respond to it while it's still on the stack and give your creature the effects from VoV.

The key idea here is that the target must be legal when the spell resolves in order for the spell to have an effect on that target. And if all of the spell's targets are illegal, the spell is countered by the game rules.

Anything you do to the target to make it illegal will counter or at least hinder the spell that targets it.
